Federico Acuna is taking the reins from Fidel Isasa in the nation's top police post.
President Fernando Lugo swore in Acuna and other high-ranking officers on Tuesday at a police academy in the Paraguayan capital of Asuncion.
Lugo's inauguration on Friday ended more than six decades of one-party rule in Paraguay. The leftist president resigned as a Roman Catholic bishop to run for office.
National police spokesman Ruben Rosas says 52 senior officers also are retiring.
